Deoxyribonuclease I, or DNase I

is used to nick DNA as a first step to incorporate labelled bases into DNA

is suitable for DNase footprinting

improves yield in primary cell isolation by removing extracellular DNA

reduces viscosity in protein samples by removal of DNA contamination

DNase I is an endonuclease that cleaves phosphodiester bonds adjacent to pyrimidines to produce polynucleotides with terminal 5′-phosphates. It preferentially cleaves double-stranded DNA and, in the presence of Mg2+ , single-stranded DNA.

Unit definition: 1 Kunitz unit catalyses an increase in absorbance of 0.001 at 260 nm per minute at 25 °C, pH 5.0, when exposed to highly polymerised calf thymus DNA (1).
