Cloning and DNA Markers

Tools that modify nucleic acids provide the foundation for many molecular biology techniques such as subcloning.  For conventional cloning of DNA fragments, we provide a variety of restriction enzymes and T4 DNA Ligase. For bacterial transformation we offer a choice of competent cells, including our unique Single Step KRX strain. 

For other cloning applications we offer T4 DNA kinase, calf alkaline phosphatase and other modifying enzymes. For RNA transcription, T7, SP6 and T3 RNA polymerases are available.

An assortment of DNA/RNA markers including bench top and step ladders enable confirmation of a successful experiment.


 

Molecular Biology Enzymes

Promega offers a variety of cloning enzymes including DNA and RNA ligases and polymerases. T7, T3, and SP6 RNA Polymerases are popular choices for in vitro transcription, as they are highly specific for their respective promoters, and produce high-quality transcripts that can later be used as templates for in vitro translation.


 

Cloning Vectors and Kits

The cloning of a specific fragment of DNA into a vector such as a plasmid is one of the foundational steps that enables additional characterization of that sequence. Vectors such as the pGEM® series contain multiple cloning sites to excerpt fragments generated by digestion with specific restriction enzymes. Sequences created by PCR and related TA cloning methods can be easily ligated into the pGEM® T-vectors. For multiple downstream applications (i.e., protein expression) the unique Flexi® Cloning System can conveniently transfer fragments to multiple vectors.


 

Molecular Weight Markers

We offer a wide variety of DNA and RNA ladders and markers for conventional gel electrophoresis applications. Many ladders are available ready-to-load, meaning loading dye is already added. Select ladders are shipped at ambient temperature, no longer requiring ice and foam coolers for transport. Broad Range Protein Molecular Weight markers are also available.
