Affinity purification is a powerful method to enrich and purify a recombinant protein from a cell lysate. Affinity tags are placed either on the N or C terminus of the protein of interest. Recombinant tagged proteins are purified from cell lysates or supernatants by selective binding, followed by washing and elution of the purified protein. After purification the protein may be cleaved with a protease to remove the affinity tag.
